If you hold your penis to tightly as you quoted,
"10 Days back i did masturbation with hand at that time to prevent sperm ejaculation i hold tightly, that time i saw few drops of blood."
It is possible you might of caused a small blood vessel to rupture.
Do you have any pain or blood after this happen or when you urinate?
If so then it needs to be checked out.
